on the spinal cholinergic and serotonergic systems. Our results demonstrate that muscarinic acetylcholine receptors, but not nicotinic acetylcholine receptors, are involved in the antiallodynic effect of EA. Specifically, spinal M 2 and M3 acetylcholine receptors, but not M1 acetylcholine receptors, were found to mediate the effect of EA. Also, spinal 5HT3 receptors, but not 5HT1A or... a significant anti-allodynic effect, which was stronger at ST36. The analgesic effect of EA at ST36 was blocked by intraperitoneal injection of muscarinic acetylcholine receptor antagonist (atropine, 1 mg/kg), but not by nicotinic (mecamylamine, 2 mg/kg) receptor antagonist. Furthermore, intrathecal administration of M2(methoctramine, 10μg) and M3 (4-DAMP, 10μg) receptor antagonist, but not M1 (pirenzepine, 10μg) receptor antagonist, blocked the effect. Also, spinal administration of 5-HT3... -

the Muscarinic-and A1-adenosine Receptor Coupled Regulation of Electrically Evoked Acetylcholine Release in the Rat Hippocampus Byung Sik Yu1’ Do Kyung Kim, and Bong Kyu Choe 2Department of Phanacology, Wonkwang University School of Medicine and Medicinal Resources Research Center of Wonkwang University, Iksan 570-749, Korea; lDepartment of Anesthesiology, College of Medicine, Chosun... It was attempted to clarify the participation of K+-channels in the post-receptor mechanisms of the muscarinic and A1-adenosine receptor- mediated control of acetylcholine (ACh) release in the present study. Slices from the rat hippocampus were equilibrated with [3H]choline and the release of the labelled products was evoked by electrical stimulation (3 Hz, 5 V/cm, 2 ms, rectangular pulses), and the influence of various agents on the evoked tritium-outflow was investigated. Oxotremorine (Oxo,... -

muscarinic receptors is not involved in determining receptor pharmacology, mutation of the sequence enhanced the coupling of M1 receptors to the stimulation of phospholipase C. Key Words: M1 muscarinic receptors, Phospholipase C, Site-mutagenesis, Carbachol INTRODUCTION Genes encoding five distinct subtypes of mam malian muscarinic acetylcholine receptors have been molecularly c10ned... alterations in the rate of receptor desensitization or sequestration. The observed larger response to carbachol at mutant M1 receptors was also not due to an artifact resulting from selection of CHO cells which express higher levels of G-proteins or phospholipase C. Our data suggest that although the LYTTYL sequence in M1 muscarinic receptors is not involved in determining receptor pharmacology, mutation of the sequence enhanced the coupling of M1 receptors to the stimulation of phospholipase C. -

muscarinic receptors to different cellular signaling pathways although they might play a role in the modulation of muscarinic receptor coupling to PI hydrolysis. Our data suggest that the LYTTYL/LYTLYT sequence differences between M1 and M2 muscarinic receptors are not important for specifying ligand binding and coupling of various subtypes of muscarinic receptors to different cellular signaling pathways although they might play a role in the modulation of muscarinic reseptor coupling to... -

receptor family members present in the intestinal mu scle is needed. Many ob- servations on the 5-HT receptor subtypes have been accu- mulated: 5-HT 1A receptors mediate nonadrenergic in- hibitory synaptic potentials in the noncholinergic secreto- motor neurons [32]. 5-HT can induce porcine proximal stomach contraction via smooth muscle 5-HT 2A receptors [33]. Acetylcholine release induced by a... concentration-dependent contractions. The 5-HT-induced contraction was enhanced by the 5HT2 receptor antagonist ketanserin and the 5-HT3 receptor antagonist ondansetron but was inhibited by the 5-HT1 receptor antagonist methysergide and 5-HT4 receptor antagonist GR113808. These results indicate that 5-HT1 and 5-HT4 receptors may mediate the contraction of the 5-HT-induced response and 5-HT2 and 5-HT3 receptors may mediate 5-HT-induced relaxation in feline ileal longitudinal smooth muscles. -

in various pathophysiologic changes [8,23]. Muscarinic and purinergic receptors are well known to raise [Ca 2+ ]i by releasing Ca 2+ from intra- cellular stores [24]. We, previously, demonstrated that the M3 and M 5 muscarinic subtypes we re expressed and mobi- lized Ca 2+ via PLC-IP3-dependent pathway in retinoblas- toma cells [25]. The involvement of PLC in the Ca 2+ re- sponses to... P2Y receptors are metabotropic G-protein-coupled receptors, which are involved in many important biologic functions in the central nervous system including retina. Subtypes of P2Y receptors in retinal tissue vary according to the species and the cell types. We examined the molecular and pharmacologic profiles of P2Y purinoceptors in retinoblastoma cell, which has not been identified yet. To achieve this goal, we used Ca2+ imaging technique and western blot analysis in WERI-Rb-1 cell, a human... -

by acetylcholine released from ex- citatory enteric motor neurons acting on muscarinic re- ceptors located in the smooth muscle [3, 28-30]. Except for acetylcholine, other neurotransmitters influence EFS-in- duced contraction in esophage al smooth muscle. In this study, we examined the role of P2X and P2Y-purinoceptors in EFS-induced contraction using cat esophageal smooth muscle. METHODS... M) also potentiated EFS-induced contractions in a dose-dependent manner. Ecto-ATPase activator apyrase (5 and 10 U/ml) reduced EFS-induced contractions. Inversely, 6-N,N-diethyl-D-Ղ,Ճ- dibromomethylene 5'-triphosphate triammonium (ARL 67156, 10−4 M) increased EFS-induced contraction. These data suggest that endogenous ATP plays a role in EFS-induced contractions which are mediated through both P2X-receptors and P2Y-receptors stimulation in cat esophageal smooth muscle.
