Effect of C1 Esterase Inhibitor on the Cardiac Dysfunction Following Ischemia and Reperfusion in the Isolated Perfused Rat Heart
Geon-YoungLee, Yong-KyooShin, Yoon-YoungJang, Jin-HoSong, Dae-JoongKim 대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 8 Pages
대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 1999, Vol.3 No.6 6 579-586 (8 pages)
Complement-mediated neutrophil activation has been hypothesized to be an important mechanism of reperfusion injury. It has been proposed that C1 esterase inhibitor (C1 INH) may prevent the complement- dependent activation of polymorphonuclear leukocytes (PMNs) that occurs within postischemic myocardium. Therefore, The effect of C1 INH was examined in neutrophil dependent isolated perfused rat heart model of ischemia (I) (20 min) and reperfusion (R) (45 min). Administration of C1 INH (5 mg/Kg)... -

This study was undertaken to examine the effect of ethanol on Na-dependent phosphate (Na-Pi) uptake in opossum kidney (OK) cells, an established renal proximal tubular cell line. Ethanol inhibited Na-dependent component of phosphate uptake in a dose-dependent manner with I50 of 8.4%, but it did not affect Na-independent component. Similarly, ethanol inhibited Na-dependent uptakes of glucose and amino acids (AIB, glycine, alanine, and leucine). Microsomal... -
Lipid Peroxidation, NF-κB Activation and Cytokine Production in Neutrophil-Stimulated Pancreatic Acinar Cells

13α-acetate (PMA) affect the productions H2O2 and lipid peroxide (LPO), NF-κB activation and cytokine production in pancreatic acinar cells, and whether these alterations were inhibited by an antioxidant, N-acetylcysteine (NAC) and superoxide dismutase (SOD). H2O2 (ferrithiocyanate method), LPO (as thiobarbiturate reactive substances), and cytokines (IL-1β, IL-6, TNF-α; enzyme-linked immunosorbent assay) and NF-κB activation (electrophoretic mobility shift assay) were analyzed in acinar... -
PAF Contributes to Intestinal Ischemia/Reperfusion-Induced Acute Lung Injury through Neutrophilic Oxidative Stress

the productions of PAF in the lung and intestine were increased (p<0.001) after intestinal I/R compared with sham rats. The level of PAF in plasma was also increased (p<0.05) following intestinal I/R. In cytochemical electron microscopy, the generation of hydrogen peroxide was increased after intestinal I/R in the lung and intestine, but decreased by treatment of WEB 2086 in the lung as well as intestine. Collectively, these experimental results indicate that PAF is the humoral mediator to... -
Reoxygenation Stimulates EDRF(s) Release from Endothelial Cells of Rabbit Aorta
SukHyoSuh, JaeJinHan, SungJinPark, JaiYoungChoi, JaeHoonSim, YoungChulKim, KiWhanKim* 대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 12 Pages
대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 1999, Vol.3 No.4 4 393-404 (12 pages)
We have reported that hypoxia stimulates EDRF(s) release from endothelial cells and the release may be augmented by previous hypoxia. As a mechanism, it was hypothesized that reoxygenation can stimulate EDRF(s) release from endothelial cells and we tested the hypothesis via bioassay experiment. In the bioassay experiment, rabbit aorta with endothelium was used as EDRF donor vessel and rabbit carotid artery without endothelium as a bioassay test ring. The test ring was contracted by... -
Effects of Carnosine and Related Compounds on Monosaccharide Autoxidation and H2O2 Formation

대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 1999, Vol.3 No.3 3 251-261 (11 pages)
The effects of carnosine and related compounds (CRCs) including anserine, homocarnosine, histidine, and β-alanine on monosaccharide autoxidation and H2O2 formation were investigated. The incubation of CRCs with D-glucose, D-glucosamine, and D, L-glyceraldehyde at 37oC increased the absorption maxima at 285 nm, 273 nm, and 290∼330 nm, respectively. D, L-glyceraldehyde was the most reactive sugar with CRCs. The presence of copper strongly stimulated the reaction of carnosine and anserine with... -
The Mechanism of t-Butylhydroperoxide-Induced Apoptosis in IMR- 32 Human Neuroblastoma Cells

대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 1999, Vol.3 No.1 3 19-28 (10 pages)
Apoptosis has been implicated in the pathophysiological mechanisms of various neurodegenerative diseases. In a variety of cell types, oxidative stress has been demonstrated to play an important role in the apoptotic cell death. However, the exact mechanism of oxidative stress-induced apoptosis in neuronal cells is not known. In this study, we induced oxidative stress in IMR-32 human neuroblastoma cells with tert- butylhydroperoxide (TBHP), which was confirmed by significantly reduced... -

histochemical electron microscopy, increased generation of oxidants was identified around neutrophils and in the cytoplasm of type II pneumocytes. Mepacrine reduced the generation of oxidants in the tissue produced by neutrophilic respiratory burst. In immunoelectron microscopic study, PLA2 was identified in the cytoplasm of the type II pneumocytes after IL-1 treatment, but mepacrine diminished PLA2 particles in the cytoplasm of the type II pneumocyte. Based on these experimental results, it is... -

The present study was undertaken to examine the role of phospholipase A2 (PLA2) in oxidant-induced inhibition of phosphate transport in primary cultured rabbit renal proximal tubule cells. Uptakes of phosphate and glucose were dose-dependently inhibited by an oxidant t-butylhydroperoxide (tBHP), and the significant inhibition appeared at 0.025 mM of tBHP, whereas tBHP-induced alterations in lipid peroxidation and cell viability were seen at 0.5 mM. tBHP stimulated arachidonic acid (AA) release... -
Protective Mechanism of Nitric Oxide and Mucus against Ischemia/ Reperfusion-Induced Gastric Mucosal Injury

Lipid peroxide production, the contents of glutathione and mucus, and glutathione peroxidase activities of gastric mucosa were determined. Histological observation of gastric mucosa was performed by using hematoxylin-eosin staining and scanning electron microscopy. The result showed that ischemia/reperfusion increased lipid peroxide production and decreased the contents of glutathione and mucus as well as glutathione peroxidase activities of gastric mucosa. Ischemia/reperfusion induced gastric...
