The Korean Journal of Physiology & Pharmacology 2017, Vol.21 No.4 10 439-447 (9 pages)
a genetic disease that displays impaired relaxation of skeletal muscle and muscle hypertrophy. This disease is mainly caused by mutations of CLCN1 that encodes human skeletal muscle chloride channel (CLC-1). CLC-1 is a voltage gated chloride channel that activates upon depolarizing potentials and play a major role in stabilization of resting membrane potentials in skeletal muscle. In this study, we report 4 unrelated Korean patients diagnosed with myotonia congenita and their clinical features.... -
JS-III-49, a hydroquinone derivative, exerts anti-inflammatory activity by targeting Akt and p38

cytokines interleukin-6 (IL-6) and IL-1β without cytotoxicity in LPS-stimulated RAW264.7 cells. JS-III-49 inhibited nuclear translocation of the NF-κB transcription factors p65 and p50 by directly targeting Akt, an upstream kinase of theNF-κB pathway, in LPS-stimulated RAW264.7 cells. However, JS-III-49 did not directly inhibit the kinase activities of Src and Syk, which are upstream kinases of Akt, in LPSstimulatedRAW264.7 cells. Moreover, JS-III-49 suppressed the nuclear translocation of... -
Chronic Ca2+ influx through voltage-dependent Ca2+ channels enhance delayed rectifier K+ currents via activating Src family tyrosine kinase in rat hippocampal neurons
The Korean Journal of Physiology & Pharmacology 7 Pages
The Korean Journal of Physiology & Pharmacology 2017, Vol.21 No.2 14 259-265 (7 pages)
Excessive influx and the subsequent rapid cytosolic elevation of Ca2+ in neurons is the major cause to induce hyperexcitability and irreversible cell damage although it is an essential ion for cellular signalings. Therefore, most neurons exhibit several cellular mechanisms to homeostatically regulate cytosolic Ca2+ level in normal as well as pathological conditions. Delayed rectifier K+ channels (IDR channels) play a role to suppress membrane excitability by inducing K+ outflow in various... -
Novel functional roles of caspase-related genes in the regulation of apoptosis and autophagy

by genetic screening of 15,000 EP lines. However, the cellular functions and molecular mechanisms of the screened genes, such as their involvement in apoptosis and autophagy, are poorly understood in mammalian cells. In order to study the functional characterizations of the genes in human cells, we investigated 16 fulllength human genes in mammalian expression vectors and tested their effects on apoptosis and autophagy in human cell lines. Our studies revealed that ALFY, BIRC4, and TAK1 induced... -
Molecular association of CD98, CD29, and CD147 critically mediates monocytic U937 cell adhesion
The Korean Journal of Physiology & Pharmacology 9 Pages
The Korean Journal of Physiology & Pharmacology 2016, Vol.20 No.5 9 515-523 (9 pages)
Adhesion events of monocytes represent an important step in inflammatory responses induced by chemokines. The β1-integrin CD29 is a major adhesion molecule regulating leukocyte migration and extravasation. Although several adhesion molecules have been known as regulators of CD29, the molecular interactions between CD29 and its regulatory adhesion molecules (such as CD98 and CD147) have not been fully elucidated. Therefore, in this study, we examined whether these molecules are functionally,... -
p53 signaling is involved in leptin-induced growth of hepatic and breast cancer cells
The Korean Journal of Physiology & Pharmacology 12 Pages
The Korean Journal of Physiology & Pharmacology 2016, Vol.20 No.5 6 487-498 (12 pages)
Leptin, an adipokine predominantly produced from adipose tissue, is well known to induce tumor growth. However, underlying molecular mechanisms are not established yet. While p53 has long been well recognized as a potent tumor suppressor gene, accumulating evidence has also indicated its potential role in growth and survival of cancer cells depending on experimental environments. In the present study, we examined if p53 signaling is implicated in leptin-induced growth of cancer cells. Herein, we... -
Dexmedetomidine inhibits vasoconstriction via activation of endothelial nitric oxide synthase
The Korean Journal of Physiology & Pharmacology 7 Pages
The Korean Journal of Physiology & Pharmacology 2016, Vol.20 No.5 1 441-447 (7 pages)
Despite the complex vascular effects of dexmedetomidine (DEX), its actions on human pulmonary resistance arteries remain unknown. The present study tested the hypothesis that DEX inhibits vascular tension in human pulmonary arteries through the endothelial nitric oxide synthase (eNOS) mediated production of nitric oxide (NO). Pulmonary artery segments were obtained from 62 patients who underwent lung resection. The direct effects of DEX on human pulmonary artery tension and changes in vascular... -

the pathway, a comprehensive approach to delineate the biological interactions of the molecules is required. However, quantitative modeling of the mitochondria-ROS generating pathway based on experiment and systemic analysis using the model have not been attempted so far. Thus, we conducted experiments to measure the concentration changes of critical molecules associated with mitochondrial apoptosis in both human mesothelioma H2052 and their ρ0cells lacking mitochondrial DNA (mtDNA). Based on... -
Resveratrol attenuates lipopolysaccharide-induced dysfunction of blood-brain barrier in endothelial cells via AMPK activation
The Korean Journal of Physiology & Pharmacology 8 Pages
The Korean Journal of Physiology & Pharmacology 2016, Vol.20 No.4 1 325-332 (8 pages)
Resveratrol, a phytoalexin, is reported to activate AMP-activated protein kinase (AMPK) in vascular cells. The blood-brain barrier (BBB), formed by specialized brain endothelial cells that are interconnected by tight junctions, strictly regulates paracellular permeability to maintain an optimal extracellular environment for brain homeostasis. The aim of this study was to elucidate the effects of resveratrol and the role of AMPK in BBB dysfunction induced by lipopolysaccharide (LPS). Exposure of... -
Klotho plays a critical role in clear cell renal cell carcinoma progression and clinical outcome

to that of adjacent non-neoplastic parenchyma. Second, the Klotho expression is clearly low in higher grade of ccRCC and is closely associated with clinical outcomes in tumor progression. Third, Klotho suppresses IGF-1-stimulated cell proliferation and migration by inhibiting PI3K/Akt pathway. These results provide compelling evidence supporting that Klotho acting on IGF-1R signaling functions as tumor suppressor in ccRCC and suggest that Klotho is a potential carcinostatis substance for ccRCC.
