The effects of pH on K currents were investigated in single smooth muscle cells isolated from the thoracic aorta of Wistar-Kyoto rats. Whole-cell K currents were recorded in the conventional configuration of the voltage-clamp technique. Pinacidil (10uM) activated the whole-cell current and the pinacidil-activated current was completely inhibited by glibenclamide (10uM) , an inhibitor of ATP-sensitive K channel (KATP channel). Pinacidil-activated current was reversed at near... -
Role of Intracellular Taurine in Monensin-induced Na+, Ca++ Accumulation and Mechanical Dysfunction in Isolated Rat Hearts

It has been postulated that the intracellular taurine is co-transported with Na down a concentration gradient and prevents the intracellular accumulation of sodium. It is therefore, expected that an elevated level of intracellular taurine prevents the sodium-promoted calcium influx to protect the cellular damages associated with sodium and calcium overload. In the present study, we evaluated the effects of intra- and extracellular taurine on the myocardial Na and Ca... -
Regulation of AQP-4 Water Channel Expression in the Brain during Development and by Ischemia

Water transport is mediated by two distinct pathways, diffusional and channel-mediated water transport. The first molecular water channel was identified from human erythrocytes in 1992. Genetically-related proteins from other mammalian tissues have subsequently been identified to transport water, and the group is referred to as th "Aquaporins". Aquaporin-4 (AQP4) is most abundant in the brain, which may be involved in CSF reabsorption and osmoregulation. However, ontogeny and regulatory... -
Effect of t-butylhydroperoxide on Na+-dependent Glutamate Uptake in Rabbit Brain Synaptosome

by an inhibition of glutamate uptake. The impairment of glutamate uptake by t-BHP was not intimately related to the change in Na+-K+-ATPase activity. These results suggest that inhibition of glutamate uptake by t-BHP is not totally mediated by peroxidation of membrane lipid, but is associated with direct interactions of glutamate transport proteins with t-BHP metabolites. The Ca2+ influx through Ca2+ channel or PLA2 activation may not be involved in the t-BHP inhibition of glutamate transport. -

We have investigated the two types of voltage-dependent outward potassium (K) currents, i.e. delayed rectifier K current (IK(V)) and 'A-like' transient outward K current (Ito) with patch-clamp technique in single smooth muscle cells (SMCs) isolated from rabbit basilar artery, and investigated the characteristics of them. The time-courses of activation were well fitted by exponential function raised to second power (n2) in IK(v) and fourth power (n4) in Ito. The activation, inactivation and... -
Inhibition of Phospholipase A2 Diminishes the Acute Alveolar Injury Induced by Interleukin-1

In an attempt to investigate the role of phospholipase A2(PLA2) in interleukin-1 (IL-1) induced acute lung injury, mepacrine was tried to inhibit PLA2 in IL-1 induced ARDS rats. For confirmation of acute lung injury by IL-1, and to know the role of neutrophils in this injury, lung leak index, lung myeloperoxidase(MPO), number of neutrophils and protein content in the bronchoalveolar lavage (BAL) and wet lung weight were measured. At the same time lung PLA2 was measured to know the effect of... -
Effect of Hydroxocobalamin on Contractile Responses to Phenylephrine during Administration of Inhalational Anesthetics in Lipopolysaccharide-Treated Rat Aortae

The hemodynamic changes in septic patients produced by inhalational anesthetics are sufficient to threaten the anesthesiologists. The effect of hydroxocobalamin, a vitamin B12a, on contractile responses to phenylephrine during administration of inhalational anesthetics were evaluated in aortic ring preparations obtained from LPS-treated rats. The sepsis was developed by intraperitoneal injection of LPS (1.5 mg/kg for l8h) and confirmed by iNOS expression using RT-PCR. Statistical significances...
