The present study was designed to examine the effect of d-amphetamine on CA release from the isolated perfused model of the rat adrenal gland, and to establish its mechanism of action. D- amphetamine (10∼100μM), when perfused into an adrenal vein of the rat adrenal gland for 60 min, enhanced the CA secretory responses evoked by ACh (5.32⁓103 M), excess K (5.6⁓102 M, a membrane depolarizer), DMPP (104 M, a selective neuronal nicotinic Nn-receptor agonist)... -

To prove the buffering contribution of mitochondria to the increase of intracellular Ca2+ level ([Ca2+]i) via background nonselective cation channel (background NSCC), we examined whether inhibition of mitochondria by protonophore carbonylcyanide m-chlorophenylhydrazone (CCCP) affects endothelial Ca2+ entry and Ca2+ buffering in freshly isolated rabbit aortic endothelial cells (RAECs). The ratio of fluorescence by fura-2 AM (R340/380) was measured in RAECs. Biological state was checked by... -

Recent data have shown the importance of oxidative stresses in the pathogenesis of inflammatory bowel disease, crohn's disease and ulcerative colitis. H2O2, reactive oxygen species (ROS) donor, has been reported to act as a signaling molecule involved in a variety of cellular functions such as apoptosis and proliferation. In the present study, we investigated viability of cultured ileal smooth muscle cells (ISMC) after stimulation with H2O2. Trypan blue method revealed that the cell viability of... -
Mechanism of Leptin-Induced Potentiation of Catecholamine Secretion Evoked by Cholinergic Stimulation in the Rat Adrenal Medulla

The aim of the present study was to examine the effect of leptin on CA release from the isolated perfused model of the rat adrenal gland, and to establish its mechanism of action. Leptin (1∼100 ng/ml), when perfused into an adrenal vein of the rat adrenal gland for 60 min, enhanced a dose-dependently the secretory responses of CA evoked by ACh (5.32⁓103 M), DMPP (104 M) and McN-A-343 (104 M), although it alone has weak effect on CA secretion. However, it did not... -

Using phospholipase D1 (PLD1)-overexpressing PC12 (PLD1-PC12) cells, the regulatory roles of PLD1 on ATP-induced currents were investigated. In control and PLD1-PC12 cells, ATP increased PLD activity in an external Ca2 dependent manner. PLD activity stimulated by ATP was substantially larger in PLD1-PC12 cells than in control cells. In whole-cell voltage-clamp mode, ATP induced transient inward and outward currents. The outward currents inhibited by TEA or charybdotoxin were significantly... -

affect basal CA output. In adrenal glands loaded with DX (3⁓107 M), CA secretory responses evoked by Bay-K-8644, an activator of L-type Ca2 channels and cyclopiazonic acid, an inhibitor of cytoplasmic Ca2-ATPase were time-dependently inhibited. Furthermore, daunorubicin (3⁓107 M), given into the adrenal gland for 60 min, attenuated CA secretory responses evoked by ACh, high K, DMPP and McN-A-343. Taken together, these results suggest that DX causes... -

smooth muscle cell derived form the circular muscle layer of the intestine, preferential hydrolysis of PIP2 and generation of IP3 and IP3-dependent Ca2 release initiate the contraction. In smooth muscle cells derived from longitudinal muscle layer of the intestine, preferential hydrolysis of PC by PLA2, generation of AA and AA-mediated Ca2 influx, cADP ribose formation and Ca2-induced Ca2 release initiate the contraction. Sustained contraction, however, in both cell... -
The Effect of Indomethacin on the Production of Eicosanoids and Edema during Ischemia-Reperfusion Injury in Skeletal Muscle

During reperfusion of skeletal muscle after ischemia, lipid mediators, mainly eicosanoids, are released and may have a role in the pathogenesis of reperfusion injury. To validate the role of eicosanoids in the ischemia-reperfusion induced functional deficits in skeletal muscle, we compared muscle edema and the changes of eicosanoid concentration in the rat hind limb after ischemia-reperfusion injury by application of tourniquet. After 4 hours of ischemia, reperfusion was established for 4 hours... -

Many reports suggest that neurotensin (NT) in the gastrointestinal tract may play a possible role as a neurotransmitter, a circulating hormone, or a modulator of motor activity. NT exerts various actions in the intestine; it produces contractile and relaxant responses in intestinal smooth muscle. This study was designed to investigate the effect of NT on motility of antral circular muscle strips in guinea-pig stomach. To assess the role of Ca2 influx in underlying mechanism, slow waves... -

basal IBa, and SIN-1 (10μM) inhibited peak IBa by 32.4⁑5.8% (n=5) in the presence of Cu/Zn SOD. In a cell clamped at a holding-potential of 40 mV, application of 10μM histamine induced an inward current. The histamine-induced inward current was markedly and reversibly inhibited by 10μM SIN-1, and this effect was abolished by ODQ (1μM). In addition, SIN-1 markedly increased the depolarization-activated outward K currents in the all potential ranges. We concluded that SIN-1...
