We investigated the action of NOS inhibitors on NOS in rats. Both of nitric oxide synthase inhibitors, NG-monomethyl-L-arginine (L-NMMA, 3μM) or NG-nitro-L-arginine methylester (L-NAME, 30μM), augmented phenylephrine (PE, 107 M)-induced contraction which was inhibited by acetylcholine (ACh) in rat thoracic aorta. This augmentation by L-NAME or L-NMMA was attenuated with the treatment of NO precursor, arginine. ACh, however, decreased the augmentation induced by L-NMMA, but not by... -

NI-RCA. Endothelium-dependent relaxation by calcium ionophore A23187 was inhibited in I-RCA and VI-RCA, and recovered in NI-RCA. Cyclic GMP contents were decreased in I-RCA group alone. Electrical field stimulation in C-RCA produced transient and frequency-dependent contractions and at 50 Hz caused biphasic contractions. The transient contractions were not affected by pretreatment with phentolamine and atropine, but the biphasic contraction was altered by the pretreatment. Both contractions were... -

This study evaluated the effects of PKC activation using phorbol 12-myristate 13-acetate (PMA) and PKC inhibition using the isoquinoline sulfomide derivative H-7 on hemodynamics and glucoregulation in the isolated perfused rat liver. Livers were isolated from fed male Holtzman rats and perfused with Krebs Ringer bicarbonate solution under a constant flow of 50 ml/min at 35oC. Portal vein pressure, glucose and lactate concentrations in the medium and oxygen consumption rates were continuously... -

that STZ-treated rats increased the maximal contractile response of the aorta to phenylephrine (PE), and high K, while the sensitivity remained unaltered. Endothelium-dependent relaxation, but not SNP-mediated relaxation, was reduced in STZ-treated rats. Plasma nitrite/nitrates are significantly increased in STZ-treated rats compared to controls. The malondialdehyde (MDA) contents of liver, serum, and aorta of diabetic rats were also significantly increased. Furthermore, nitrotyrosine, a... -
Characteristics of Hypoxic Pulmonary Vasoconstriction of the Rat Study by the Vessel Size and Location in the Lung

Pulmonary blood vessels with diameters of 200∼400 μm produce considerably more force in response to vasoconstrictor drugs than those which are either smaller or larger. We have therefore investigated whether or not hypoxic pulmonary vasoconstriction (HPV) is more powerful in vessels of these diameters. We have also looked at the possibility that vessels from different regions of the lung respond differently. To do this we have grouped vessels according to their location within the lung as... -
Evidence for Adenosine Triphosphate (ATP) as an Excitatory Neurotransmitter in Guinea-Pig Gastric Antrum

We explore the question of whether adenosine 5'-triphosphate (ATP) acts as an excitatory neurotransmitter in guinea-pig gastric smooth muscle. In an organ bath system, isometric force of the circular smooth muscle of guinea-pig gastric antrum was measured in the presence of atropine and guanethidine. Under electrical field stimulation (EFS) at high frequencies (>20 Hz), NO-mediated relaxation during EFS was followed by a strong contraction after the cessation of EFS (a... -
Roles of Na+-Ca2+ Exchange in the Negative Force-Frequency Relationship

Frequency-force relationships (FFR) were studied in electrically field stimulated rat left atria (LA) by reducing the stimulation frequency from resting 3 Hz to test frequencies (0.1-1 Hz) for 5 minutes. The twitch amplitudes of LA elicited the typical negative staircases with 3-phased changes: the initial rapid increase, the second decrease and the following plateau at test frequencies. Verapamil (3⁓105 M) pretreatment elicited frequency-dependent suppression of the twitch... -
Moderate Elevation of Extracellular K+ Concentration Induces Vasorelaxation in Isolated Rat, Rabbit and Human Cerebral Arteries Role of Na Pump and Ba-Sensitive Process

in the rabbits and induced relaxation in the rats and humans. Addition of K (2∼10 mM) to the control solution induced vasorelaxations. These effects were inhibited by the pretreatment with both ouabain (10 ㄍM) and Ba2 (0.1∼0.3 mM) in the rat, but only with ouabain (10 ㄍM) in the rabbit and human. These results suggest that K-induced vasorelaxation occurs via the stimulation of electrogenic Na pump in the rabbit and human MCAs, while in the rat MCAs via the... -

6 mM to 12 mM. K-induced relaxation was evoked with the stepwise increment of extracellular K from 0 or 2 mM to 12 mM by 2 mM in basilar arterial rings, which were contracted by histamine. But [K]O elevation from 4 or 6 mM to 12 mM by the stepwise increment evoked no significant relaxation. Basal tension of basilar artery was increased with [K]O elevation from 6 mM to 12 mM by 2 mM steps or by the treatment with ouabain and the increase of basal tension was blocked by...
