Effects of Cl Channel Blockers on the Cardiac ATP-sensitive K+ Channel
The Korean Journal of Physiology & Pharmacology 9 Pages
The Korean Journal of Physiology & Pharmacology 1999, Vol.3 No.3 8 305-313 (9 pages)
DIDS blocked the pinacidil- or 2,4-dinitrophenol (DNP)-induced KATP channel openings. In contrast, niflumic acid did not block the pinacidil-induced KATP channel openings in inside-out patches, but inhibited it in cell-attached patches. DIDS and niflumic acid produced additional block in the presence of ATP and did not affect current-voltage relationship and channel kinetics. All these results indicate that DIDS among Cl channel blockers specifically blocks the cardiac KATP channel. -
Opening of ATP-sensitive K+ Channel by Pinacidil Requires Serine/ Threonine Phosphorylation in Rat Ventricular Myocytes
The Korean Journal of Physiology & Pharmacology 11 Pages
The Korean Journal of Physiology & Pharmacology 1999, Vol.3 No.3 7 293-303 (11 pages)
The influences of specific protein phosphatase and protein kinase inhibitors on the ATP-sensitive K (KATP) channel-opening effect of pinacidil were investigated in single rat ventricular myocytes using patch clamp technique. In cell-attached patches, pinacidil (100 μM) induced the opening of the KATP channel, which was blocked by the pretreatment with H-7 (100 μM) whereas enhanced by the pretreatment with genistein (30 μM) or tyrphostin A23 (10 μM). In inside-out patches, pinacidil... -
Thyroid Hormone-Induced Alterations of Ca2+-ATPase and Phospholamban Protein Expression in Cardiac Sarcoplasmic Reticulum
The Korean Journal of Physiology & Pharmacology 8 Pages
The Korean Journal of Physiology & Pharmacology 1999, Vol.3 No.2 13 223-230 (8 pages)
Alterations of cardiovascular function associated with various thyroid states have been studied. In hyperthyroidism left ventricular contractility and relaxation velocity were increased, whereas these parameters were decreased in hypothyroidism. The mechanisms for these changes have been suggested to include alterations in the expression and/or activity levels of various proteins; α-myosin heavy chain, β- myosin heavy chain, β-receptors, the guanine nucleotide-binding regulatory protein, and... -
Effects of Ethanol on Na+-dependent Solute Uptake in Rabbit Renal Brush-Border Membrane Vesicles
The Korean Journal of Physiology & Pharmacology 8 Pages
The Korean Journal of Physiology & Pharmacology 1999, Vol.3 No.2 9 191-198 (8 pages)
This study was undertaken to examine the effect of ethanol on Na-dependent transport systems (glucose, phosphate, and dicarboxylate) in renal brush-border membrane vesicles (BBMV). Ethanol inhibited Na- dependent uptakes of glucose, phosphate, and succinate in a dose-dependent manner, but not the uptakes of Na-independent. The H/TEA antiport was reduced by 8% ethanol. Kinetic analysis showed that ethanol caused a decrease in Vmax of three transport systems, leaving... -
Properties of Spontaneous Activity in Gastric Smooth Muscle
H.Suzuki, Y.Yamamoto, G.D.S.Hirst 대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 7 Pages
The Korean Journal of Physiology & Pharmacology 1999, Vol.3 No.2 1 119-125 (7 pages)
Mammalian gastric smooth muscles generate spontaneous rhythmic contractions which are associated with slow oscillatory potentials (slow waves) and spike potentials. Spike potentials are blocked by organic Ca2-antagonists, indicating that these result from the activation of L-type Ca2-channel. However, the cellular mechanisms underlying the generation of slow wave remain unclear. Slow waves are insensitive to Ca2-antagonists but are blocked by metabolic inhibitors or low... -
Moderate Elevation of Extracellular K+ Concentration Induces Vasorelaxation in Isolated Rat, Rabbit and Human Cerebral Arteries Role of Na Pump and Ba-Sensitive Process
The Korean Journal of Physiology & Pharmacology 10 Pages
The Korean Journal of Physiology & Pharmacology 1998, Vol.2 No.6 7 705-714 (10 pages)
Cerebral blood vessels relax when extracellular K concentrations ([K])e are elevated moderately (2∼15 mM, K-induced vasorelaxation). We have therefore studied the underlying mechanism for this K- induced vasorelaxation in the isolated middle cerebral arteries (MCAs). The effects of ouabain and Ba2 on K-induced vasorelaxation were examined to determine the role of sodium pump and/or Ba-sensitive process (possibly, inward rectifier K current) in the... -
Altered Cerebral Vasomotion with Decreased CGRP Level in Pial Arteries of Spontaneously Hypertensive Rats
The Korean Journal of Physiology & Pharmacology 8 Pages
The Korean Journal of Physiology & Pharmacology 1998, Vol.2 No.5 5 573-580 (8 pages)
contrast, SDR and WR showed a large increase in diameter, and the increase was sustained for over 10 minutes. In line with these results, the basal releases of CGRP-like immunoreactivity (CGRP-LI) in the isolated pial arteries from SHR with high and normotensive BP were 12.5⁑1.4 and 9.8⁑2.8 fmole/mm2/60 min (P<0.05), while those from SDR and WR were 25.5⁑3.1 and 24.6⁑3.1 fmole/mm2/60 min, respectively. The isolated basilar arteries showed similar results to those of the... -
Dual Action of d-Tubocurarine on Large-Conductance Ca2+-activated K+ Channels from Rat Brain Reconstituted into Planar Lipid Bilayer
The Korean Journal of Physiology & Pharmacology 5 Pages
The Korean Journal of Physiology & Pharmacology 1998, Vol.2 No.5 2 549-553 (5 pages)
Ca2+-activated K+ channel from rat brain. When the initial open probability (Po) of the channel was relatively high, dTC decreased channel activity in a concentration dependent manner. In contrast, when the initial Po was lower, sub-micro molar dTC increased channel activity by destabilizing the closed states of the channel. Further addition of dTC up to micro molar range decreased channel activity. This dual effect of dTC implicates that there exist at least two different binding sites for dTC. -
Increased Activity of Large Conductance Ca2+-Activated K+ Channels in Negatively-Charged Lipid Membranes
The Korean Journal of Physiology & Pharmacology 11 Pages
The Korean Journal of Physiology & Pharmacology 1998, Vol.2 No.4 15 529-539 (11 pages)
The effects of membrane surface charge originated from lipid head groups on ion channels were tested by analyzing the activity of single large conductance Ca2-activated K (maxi K) channel from rat skeletal muscle. The conductances and open-state probability (Po) of single maxi K channels were compared in three types of planar lipid bilayers formed from a neutral phosphatidylethanolamine (PE) or two negatively-charged phospholipids, phosphatidylserine (PS) and phosphatidylinositol... -
Inducible Nitric Oxide Synthase mRNA Expression and Nitric Oxide Production in Silica-Induced Acute Inflammatory Lung Injury
The Korean Journal of Physiology & Pharmacology 7 Pages
The Korean Journal of Physiology & Pharmacology 1998, Vol.2 No.2 12 233-239 (7 pages)
Stimulated alveolar macrophages and neutrophils produce nitric oxide, a free radical by an inducible nitric oxide synthase(iNOS), which reacts with superoxide anion to form peroxynitrite, a more highly reactive toxic species. The objectives of the present study were to evaluate acute inflammatory lung injury and to determine iNOS mRNA induction and nitric oxide production by rat broncho-alveolar lavage cells following intratracheal treatment of silica. After 4 h exposure to silica, differential...
