the importance of oxidative stresses in the pathogenesis of inflammatory bowel disease, crohn's disease and ulcerative colitis. H2O2, reactive oxygen species (ROS) donor, has been reported to act as a signaling molecule involved in a variety of cellular functions such as apoptosis and proliferation. In the present study, we investigated viability of cultured ileal smooth muscle cells (ISMC) after stimulation with H2O2. Trypan blue method revealed that the cell viability of ISMC treated with 1 mM... -
Cooperation of Gβ and Gαq Protein in Contractile Response of Cat Lower Esophageal Sphincter (LES)

Ach-induced contraction was antibody concentration dependent. The combination with PLC-β1 and PLC-β3 antibody completely abolished the contraction, suggesting that PLC-β1 and PLC-β3 have a synergism to inhibit the contraction in LES. PLC-β1, -β3 or -γ1 antibody did not reduce the contraction of LES cells in response to DAG (106 M), suggesting that this isozyme of PLC may not activate PKC. When Gq/11 antibody was incubated, the inhibitory effect of the incubation of PLC β3, but not... -
Antagonists of NMDA Receptor, Calcium Channel and Protein Kinase C Potentiate Inhibitory Action of Morphine on Responses of Rat Dorsal Horn Neuron

single iontophoretic application of morphine or an antagonist alone induced weak inhibition of wide dynamic range (WDR) cell responses to iontophoretically applied NMDA and C-fiber stimulation. Although there was a little difference in the potentiating effects, the antinociceptive action of morphine was potentiated when morphine was iontophoretically applied together with Mn2, verapamil, AP5, Mg2 or H-7. However, the potentiating action between morphine and each antagonist was not... -

a marked and continuous pressor effect (icv KH-pressor effect), and the icv pressor effect was not repeatable. However, intravenous (iv) injection of the same dose of KH induced a prominent depressor effect (iv KH-depressor effect). The icv KH-pressor effect was inhibited by acid-denaturation, while the iv KH-depressor effect was not. Simultaneous icv administration of the three agents (ouabain, diltiazem, or bumetanide: 10μg/kg each) significantly reduced the pressor effect. The icv KH-pressor... -
A Role of Mitogen Activated Protein Kinases and Inflammatory Responses in Gender Differences in Kidney Ischemia Injury

female mice to kidney ischemic injury, and if so, whether it is due to differences in mitogen activated protein kinases (MAPKs) or inflammatory responses to ischemia. Female mice were protected against kidney ischemia when compared with males. Thirty minutes of bilateral ischemia resulted in marked functional and morphological damages in males, but not in females. The ischemia-induced phosphorylation of c-jun N-terminal stress-activated protein kinases (JNKs) was higher in males than in females.... -
The Effect of Carbon Monoxide on Contraction, Cytosolic Ca2+ Level and Ionic Currents in Guinea Pig Ileal Smooth Muscle

KCl-induced contraction and this effect was blocked by ODQ (1μM), a soluble guanylyl cyclase (sGC) inhibitor. CO inhibited the 40 mM KCl-induced contraction without changing [Ca2]i. Cumulative addition of KCl induced a graded increase in [Ca2]i and muscle tension. In the presence of CO, cumulative addition of KCl induced smaller contraction than in the absence of CO. On the other hand, the increase in [Ca2]i induced by cumulative addition of KCl was only slightly decreased... -
Stationary Outward and Transient Ca2+-Dependent Currents in Hamster Oocytes

a transient outward component (Ito) and a steady-state outward component (I∞). Ito was transiently followed by the cessation of inward Ca2 current (ICa), and its current-voltage (I-V) relation was a mirror image of that of ICa. Either blockade of ICa by Co2 or replacement of Ca2 with Sr2 abolished Ito without change in I∞. Intracellular EGTA (10 mM) inhibited Ito but not I∞. suggesting strongly that generation of Ito requires intracellular Ca2. Apamin (1 nM)... -
Changes of the Level of G Protein α-subunit mRNA by Withdrawal from Morphine and Butorphanol

Specifically, the level of Gαs mRNA was decreased in the cortex and cerebellar granule layer during the morphine and butorphanol withdrawal. The level of Gαi mRNA was decreased in the dentate gyrus and cerebellar granule layer during the morphine withdrawal. However, the level of Gαi mRNA was significantly elevated during the butorphanol withdrawal. These results suggest that region-specific changes of G protein α-subunit mRNA were involved in the withdrawal from morphine and butorphanol. -

in the presence of 10 μM Cu (II). The presence of human serum albumin (HSA) decreased their effect on H2O2 formation. Carnosine and anserine has a biphasic effect on α-ketoaldehyde formation from glucose autoxidation. CRCs inhibited glycation of HSA as determined by hydroxymethyl furfural, lysine residue with free ε-amino group, and fructosamine assay. These results suggest that CRCs may be protective against diabetic complications by reacting with sugars and protecting glycation of protein. -

which the upper and lower motor neurons are progressively destroyed. Recent evidences support the role of autoimmune mechanisms in the pathogenesis of ALS. This study investigated the effects of sera from ALS patients on neuromuscular transmission in phrenic nerve-hemidiaphragm preparations and on calcium currents of single isolated dorsal root ganglion (DRG) cells in mice. Mice were injected with either control sera from healthy adults or ALS sera from 18 patients with ALS of sporadic form, for...
