of the cells with amiloride, nifedipine, ryanodine, and thapsigargin. GA dose-dependently increased intracellular Ca2+ concentration, which was attenuated by nifedipine, nimodipine, ryanodine, and thapsigargin. However, RP-cAMP, chelerythrine, tyrphostin A23, or phenylarsine oxide did not induced any significant change in GA-stimulated increase of intracellular Ca2+ concentration. These results suggest that gap junction plays an important role on the regulation of renin release and intracellular... -

s. Fluorescence intensity of both Rhod-2 AM and TMRE were initially decreased by DNP, coincident with the initial increase of Fluo-4 AM fluorescence intensity. When sarcoplasmic reticulum (SR) Ca2 was depleted by 1μM thapsigargin plus 10μM ryanodine, the initial increase of Fluo-4 AM fluorescence intensity was unaffected, however, the subsequent progressive increase was abolished. KB-R7943 delayed both the first and the second phases of cytosolic Ca2 overload, while Na-fr... -
The Effect of External Divalent Cations on Intestinal Pacemaking Activity

Electrical rhythmicity in the gastrointestinal (GI) muscles is generated by pacemaker cells, known as interstitial cells of Cajal (ICC). In the present study, we investigated the effect of external divalent cations on pacemaking activity in cultured ICC from murine small intestine by using whole-cell patch clamp techniques. ICC generated pacemaker currents under a voltage clamp or electrical pacemaker potentials under a current clamp, and showed a mean amplitude of 500⁑50 pA or... -
Inhibition of Sarcoplasmic Reticulum Ca2+ Uptake by Pyruvate and Fatty Acid in H9c2 Cardiomyocytes: Implications for Diabetic Cardiomyopathy

High extracellular glucose concentration was reported to suppress intracellular Ca2 clearing through altered sarcoplasmic reticulum (SR) function. In the present study, we attempted to elucidate the effects of pyruvate and fatty acid on SR function and reveal the mechanistic link with glucose-induced SR dysfunction. For this purpose, SR Ca2-uptake rate was measured in digitonin-permeabilized H9c2 cardiomyocytes cultured in various conditions. Exposure of these cells to 5 mM... -
Na+-Ca2+ Exchange Curtails Ca2+ before Its Diffusion to Global Ca2+i in the Rat Ventricular Myocyte

Na-Ca2 exchange (NCX) is the major Ca2 extrusion mechanism. NCX has been considered as a relaxation mechanism, as it reduces global [Ca2]i raised during activation. However, if NCX locates in the close proximity to the ryanodine receptor, then NCX would curtail Ca2 before its diffusion to global Ca2i. This will result in a global [Ca2]i decrease especially during its ascending phase rather than descending phase. Therefore, NCX would decrease the... -
Ca2+-induced Ca2+ Release from Sarcoplasmic Reticulum Negatively Regulates Myocytic ANP Release in Beating Rabbit Atria

Thapsigargin attenuated ryanodine-induced decrease in atrial dynamic changes. Blockade of L-type Ca2 channel with nifedipine abolished ryanodine-induced increase in ANP release (0.69⁑5.58% vs. 27.31⁑3.51%; P<0.001). In the presence of thapsigargin and ryanodine, nifedipine increased ANP release and decreased atrial dynamics. These data suggest that Ca2-induced Ca2 release from the SR is inversely involved in the regulation of atrial myocytic ANP release. -

Very little research has been carried out on safflower seed for the prevention and treatment of the bone deficiency diseases, including osteoporosis, which are supported by scientific evidences. In the present study, 3μl of 0.1% dried crude extract or 2μl of 0.1% dried aqueous fraction were shown to significantly accelerate the rate of differentiation of osteoblast. Also, the crude extract and aqueous fraction increased the [Ca2]i of the cultured osteoblast cells: 3μl of 0.1% dried... -
Buffering Contribution of Mitochondria to the [Ca2+]i Increase by Ca2+ Influx through Background Nonselective Cation Channels in Rabbit Aortic Endothelial Cells

To prove the buffering contribution of mitochondria to the increase of intracellular Ca2+ level ([Ca2+]i) via background nonselective cation channel (background NSCC), we examined whether inhibition of mitochondria by protonophore carbonylcyanide m-chlorophenylhydrazone (CCCP) affects endothelial Ca2+ entry and Ca2+ buffering in freshly isolated rabbit aortic endothelial cells (RAECs). The ratio of fluorescence by fura-2 AM (R340/380) was measured in RAECs. Biological state was checked by... -

into a huge inward current of 12.9⁑0.5 pA/pF. From the inward current, two different inward INCXs were identified. One was 10 μM ryanodine-sensitive, constituting 14.2⁑2.3%. It was completely blocked by CdCl2 (0.1 mM and 0.5 mM) and by Na-depletion. The other was identified by 5 mM NiCl2 after suppression of ICaL and ryanodine receptor, constituting 14.8⁑1.6%. This latter was blocked by either 10 mM caffeine-induced SR Ca2-depletion or 1 mM tetracaine.... -

ryanodine receptor) mRNA was about 50∼60% at birth and increased gradually to adult level throughout the postnatal rat heart development. 3[H]ryanodine binding increased gradually during postnatal myocardial development, which was closely correlated with ryanodine mRNA expression levels during the development except the ryanodine mRNA level at birth. These findings indicate that cardiac SR Ca2- ATPase, phospholamban, and Ca2 release channel are expressed coordinately, which may be...
