Nitric Oxide Modulation of GABAergic Synaptic Transmission in Mechanically Isolated Rat Auditory Cortical Neurons
Jong-JuLee 대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 7 Pages
대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 2009, Vol.13 No.6 9 461-467 (7 pages)
The auditory cortex (A1) encodes the acquired significance of sound for the perception and interpretation of sound. Nitric oxide (NO) is a gas molecule with free radical properties that functions as a transmitter molecule and can alter neural activity without direct synaptic connections. We used whole-cell recordings under voltage clamp to investigate the effect of NO on spontaneous GABAergic synaptic transmission in mechanically isolated rat auditory cortical neurons preserving functional... -

inhibitor, and ODQ, a guanylate cyclase inhibitor, did not inhibit the effects of EGCG. EGCG-induced effects on pacemaker currents were not inhibited by glibenclamide, an ATP-sensitive K+ channel blocker and TEA, a Ca2+-activated K+ channel blocker. Also, we found that EGCG inhibited the spontaneous [Ca2+]i oscillations in cultured ICC. In conclusion, EGCG inhibited the pacemaker activity of ICC and reduced [Ca2+]i oscillations by cAMP-, cGMP-, ATP-sensitive K+ channel-independent manner. -

myocytes of rabbit were isolated using proteolytic enzymes including collagenase and papain. Inside-out, attached, and whole-cell configurations were examined using the patch clamp technique. The applications of 10ՌM norepinephrine or 70 mM KCl induced tonic contractions, and 10ՌM testosterone (pharmacological concentration) evoked dose-dependent relaxations of these precontracted strips. Various K+ channel blockers, such as tetraethylammonium (TEA; 10 mM), iberiotoxin (0.1ՌM),... -

evaluating both cerebral blood flow and the plasticity of the posterior communicating artery (PcomA). Male C57BL/6 mice were anesthetized with halothane and subjected to bilateral occlusion of the common carotid artery (BCCAO) for 30 min. The regional cerebral blood flow (rCBF) was measured by laser Doppler flowmetry. The plasticity of PcomA was visualized by intravascular perfusion of India ink solution. When animals had the residual cortical microperfusion less than 15% as well as the smaller... -
Effects of Phenylephrine on the Excitability of Medial Vestibular Nuclear Neurons in Rats

Coeruleo-vestibular pathway which connects locus coeruleus and vestibular nuclei is noradrenergic. This study was designed to elucidate the effects of phenylephrine on the spontaneous activity of acutely isolated medial vestibular nuclear neurons of rat by whole-cell patch-clamp technique. Sprague-Dawley rats, aged 14 to 16 days, were used. After enzymatic digestion, dissociated medial vestibular neurons were transferred to a recording chamber mounted on an inverted microscope, and spontaneous... -
Effects of Zinc on Spontaneous Miniature GABA Release in Rat Hippocampal CA3 Pyramidal Neurons

(30μM) increased GABAergic mIPSC frequency without affecting mIPSC amplitude, but it decreased both mIPSC frequency and amplitude at higher concentrations (≥300μM). In contrast, Zn2 (3 to 100μM) did not affect glutamatergic mEPSCs, although it slightly decreased both mIPSC frequency and amplitude at 300μM concentration. Facilitatory effect of Zn2 on GABAergic mIPSC frequency was occluded either in Ca2-free external solution or in the presence of 100μM 4-aminopyridine,... -
Fluoxetine Modulates Corticostriatal Synaptic Transmission through Postsynaptic Mechanism

that the inhibitory effect of fluoxetine on sEPSC amplitude (93.0⁑1.9%, n=8) was significantly blocked, but not serotonin (84.3⁑1.6%, n=4), when the brain slice was incubated with p-chloroamphetamine (10μM), which depletes serotonin from the axon terminals and blocks its reuptake. These results suggest that fluoxetine inhibits corticostriatal synaptic transmission through postsynaptic, and that these effects are exerted through both serotonin dependent and independent mechanism. -
Higher Expression of TRPM7 Channels in Murine Mature B Lymphocytes than Immature Cells

of MIC was further increased, and the relation between current and voltage became linear under divalent cation-free conditions, demonstrating typical properties of the TRPM7. The stimulation of B cell receptors (BCR) by ligation with antibodies did not change the amplitude of MIC current. Also, increase of extracellular [Mg2]c to enhance the Mg2 influx did not affect the BCR ligation-induced death of WEHI-231 cells. Although the level of TRPM7 was not directly related with the cell... -
Acetylcholine Induces Hyperpolarization Mediated by Activation of K(Ca) Channels in Cultured Chick Myoblasts
DoyunLee, JaeheeHan, Jae-YongPark 대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 8 Pages
대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 2005, Vol.9 No.1 6 37-44 (8 pages)
Our previous report demonstrated that chick myoblasts are equipped with Ca2-permeable stretch- activated channels and Ca2-activated potassium channels (KCa), and that hyperpolarization-induced by KCa channels provides driving force for Ca2 influx through the stretch-activated channels into the cells. Here, we showed that acetylcholine (ACh) also hyperpolarized the membrane of cultured chick myoblasts, suggesting that nicotinic acetylcholine receptor (nAChR) may be another... -
Effects of PCB Congeners in Rodent Neuronal Cells in Culture

We attempted to analyze the mechanism of polychlorinated biphenyl (PCB)-induced neurotoxicity and identify the target molecules in the neuronal cells for PCBs.Since the developing neuron is particularly sensitive to PCB-induced neurotoxicity, we isolated cerebellar granule cells derived from 7-day old Sprague Dawley (SD) rats and grew cells in culture for additional 7 days to mimic PND-14 conditions. Only non-coplanar PCBs at a high dose showed a significant increase of total protein kinase C...
