NO production, iNOS mRNA expression and NF-κB binding activity. In contrast, PKC inhibitors such as H-7, Ro-318220 and staurosporine did not show any effect on them. In addition, transfection of RAW 264.7 cells with iNOS promoter linked to a CAT reporter gene revealed that tyrphostin inhibited the iNOS promoter activity through the NF-κB binding site, whereas PKC inhibitors did not. Taken together, these suggest that PTK, but not PKC pathway, is involved in the regulation of the iNOS gene... -

CREB (cyclicAMP responsible element binding protein) proteins. The KA, NMDA, and glutamate did not affect forskolin- induced increase of ERK and CREB phosphorylation. The KA decreased PMA-induced increase of phosphorylation of ERK and CREB proteins, whereas glutamate and NMDA did not affect the phosphorylation of ERK and CREB proteins induced by PMA. These findings suggest that, in C6 glioma cells, c-fos mRNA induction induced by EAAs is not mediated by phosphorylation of ERK and CREB proteins. -

at PM, LYSO, HDM and LDM were 37.7⁑12.0%, 42.4⁑12.1%, 19.2⁑5.0% and 0.7⁑1.2% in the absence of insulin. Total GLUT2 distribution in the presence of insulin was almost same as that in the absence of insulin. Present data with previous findings suggest that GLUT4 translocation may be attributed to GLUT4 phosphorylation by PKC-ζ but GLUT2 does not translocate because GLUT2 is not phosphorylated by the kinase. Therefore, GLUT phosphorylation may be required in GLUT... -
Asymmetrical Distribution of P2Y Nucleotide Receptors in Rabbit Inner Medullary Collecting Duct Cells

predominantly on the basolateral membrane. These nucleotides stimulated intracellular cAMP production with an asymmetrical profile, which was comparable to that in the stimulation of Isc. Our results suggest that the adenine and uracil nucleotides can interact with different P2Y nucleotide receptors that are expressed asymmetrically on the apical and basolateral membranes of the rabbit IMCD cells, and that both cAMP- and Ca2-dependent signaling mechanisms underlie the stimulation of Isc. -

in a dose-dependent manner. The half-maximal inhibition dose (ED50) was 48.6⁑8.4μM and the Hill coefficient was 0.73⁑0.11. Single channel conductance calculated from the slope of the relationship between single channel current and pipette potential (20∼100 mV) was not significantly altered by propofol (control: 60.0⁑2.7 pS, 0.1 mM propofol: 58.7⁑3.5 pS). However, mean closed time was surely increased. Above results indicate that propofol blocks the KATP... -
Responses of Dorsal Horn Neurons to Peripheral Chemical Stimulation in the Spinal Cord of Anesthetized Cats

neurons to mechanical stimuli increased at large and the RFs were expended, suggesting development of hypersensitization (formalin 6/10, capsaicin 8/11, and ATP 15/19, respectively). These results are suggested that formalin stimulates peripheral nociceptor, local inflammation and involvement of central sensitization, capsaicin induces central sensitization as well as affects the peripheral C-polymodal nociceptors and neurogenic inflammation, and ATP directly stimulates peripheral nociceptors. -

increase in spike frequency but did not induce bursting activity. By contrast, apamin (300 nM) selectively abolished the delayed slow AHP and induced bursting activity in type B cells. Apamin had no effect on the spike frequency of type A cells. These data suggest that there are differential roles of apamin and CTX sensitive potassium conductances in spontaneous firing patterns of MVN neurons, and these conductances are important in regulating the intrinsic rhythmicity and excitability. -

ionic currents using whole cell voltage clamp technique in rabbit cardiomyocytes and on the intracellular Ca2 transient using calcium sensitive fluorescence dye, indo-1AM. Simultaneously with recording intracellular Ca2 transients, cell contractility was estimated from the changes in length of the electrical stimulated rat cardiac myocytes. L-type Ca2 current decreased by bradykinin at the entire voltage range. Inward tail current increased initially up to its maximum about... -

in F, in normalized units) and LF/HF (3.10⁑0.57 in NF, 3.00⁑1.08 in F) and decreased HFn powers. Ethanol ingestion increased LFn and LF/HF and decreased HFn. PS after ethanol resulted in higher LFn and LF/HF and lower HFn than the prior PS. F showed a greater and more sustained HRV change than NF after ethanol. In conclusion, PS or ethanol ingestion increased LFn and LF/HF and decreased HFn. Flushed subjects showed an accentuated HRV response to ethanol. -

EGTA, the RMP was depolarized and did not show STHPs. The membrane currents showed no STOCs but only showed slowly inactivating outward currents. External TEA (1 mM) reversibly inhibited the STHPs, depolarized the RMP, reduced the membrane currents, abolished STOCs, and decreased the open probability of single KCa channel. When KV currents were isolated, the application of 4-AP (5 mM) depolarized the RMP. The important aspect of our results is that KCa channel is responsible for the generation...
