Molecular Biological Diagnosis of Meloidogyne Species Occurring in Korea
The Plant Pathology Journal 9 Pages
8 247-255 (9 pages)
(Polymerase Chain Reaction-Restriction Fragment Length Polymorphism), SCAR (Sequence Characterized Amplified Region) PCR and RAPD (Randomly Amplified Polymorphic DNA) were also utilized to develop methods for the accurate and rapid species identification of the root-knot nematode species. In the sequence analysis of the D3 expansion segment, only a few nucleotide sequence variations were detected among M. incognita, M. arenaria, and M, javanica, but not M. hapla. As a result of our haplotype... -
Multiplex RT-PCR Assay for the Detection of Apple stem grooving virus andApple chlorotic Leaf spot virus in Infected Korean Apple Cultivars
The Plant Pathology Journal 6 Pages
12 168-173 (6 pages)
RNA polymerase of Apple stem grooving virus (ASGV) and coat protein of Apple chlorotic leaf spot virus (ACLSV) were characterized from the infected apple cultivars in Korea. Based on the nucleotide sequences of the characterized partial genes, the virus gene-specific primers were designed for the detection of ASGV and ACLSV infected in species of Malus. The RT-PCR using the primers for the genes of ASGV and ACLSV successfully gave rise to 404 and 566 bp DNA fragments, respectively. Using those... -
Effect of Temperature on the Concentration of Chrysanthemum stunt viroid inCSVd-infected Chrysanthemum
The Plant Pathology Journal 3 Pages
8 152-154 (3 pages)
CSVd-infected chrysanthemum plants grown under 10oC or 15oC growth chamber for 2 months resulted a higher dilution endpoint of template RNA for Reverse transcription and polymerase chain reaction (RT-PCR) than those grown for 1 month: 10-4 (1.35 -
Biological Control of a Severe Viral Strain Using a Benign Viral Satellite RNAAssociated with Cucumber mosaic virus
The Plant Pathology Journal 8 Pages
5 131-138 (8 pages)
stunting and mosaic symptoms with one strain designated KU2, and almost symptomless with the other strain designated KU1. A satellite RNA 5 (sat-RNA) was found to be associated with the KU1 strain and was characterized as a benign viral satellite RNA. Using reverse transcription and polymerase chain reaction (RT-PCR) with sat- RNA specific primers, an amplified PCR product of about 160bp was determined and analyzed by gel electrophoresis. This naturally occurring benign viral satellite RNA was... -
RT-PCR Detection of dsRNA Mycoviruses Infecting Pleurotus ostreatus and Agaricus blazei Murrill
The Plant Pathology Journal 6 Pages
7 343-348 (6 pages)
ostreatus (isolates ASI2596, ASI2597, and Bupyungbokhoe) and Agaricus blazei Murrill were determined and compared with those of the other dsRNA mycoviruses. Partial nucleotide sequences of the purified dsRNA from ASI2596 and ASI2597 revealed RNA-dependent RNA polymerase sequences that are closely related to Oyster mushroom isometric virus 2, while nucleotide sequences and the deduced amino acid sequence from dsRNA mycovirus infecting Agaricus blazei did not show any significant homology to the... -
Symptom Determinant as RNA3 of Lily Isolates of Cucumber mosaic virus on Zucchini Squash
The Plant Pathology Journal 8 Pages
9 212-219 (8 pages)
reverse-transcriptase polymerase chain reaction (RT-PCR) using primers specific to Cucumovirus genus namely, LK-CMV, LK4-CMV, and LK5-CMV. Restriction enzymes patterns of the RTPCR products revealed that the lily isolates belonged to subgroup IA of CMV. In terms of biological properties, the lily isolates have highly similar but distinct pathogenicity as reported in other lily strains and ordinary strains of CMV. To characterize the molecular properties, cDNAs containing coat protein (CP) gene... -
Characterization and RT-PCR Detection of dsRNA Mycoviruses from the Oyster Mushroom, Pleurotus ostreatus
The Plant Pathology Journal 6 Pages
7 200-205 (6 pages)
the purified dsRNA from ASI2223 and Suhan showed RNA-dependent RNA polymerase sequences that are closely related to those of partitiviruses, including Fusarium poae virus 1, Fusarium solani virus, Rhizoctonia solani virus, Discula destructiva virus 2, and Oyster mushroom isometric virus 2. Specific primers were designed for RT-PCR detection of dsRNA viruses from the P. ostreatus isolate ASI2223 and Suhan. Two virus specific primer sets were found to specifically detect each virus among six sets... -
Improvement of RT-PCR Sensitivity for Fruit Tree Viruses by Small-scale dsRNA Extraction and Sodium Sulfite
The Plant Pathology Journal 5 Pages
12 142-146 (5 pages)
of reverse transcriptase and/or Taq polymerase in RT-PCR. The commonly used multiplestep protocols using several additives to diminish polyphenolic compounds during nucleic acid extraction are time consuming and laborious. In this study, sodium sulfite was evaluated as an additive for nucleic acid extraction from woody plants and the efficiency of RT-PCR assay of commercial nucleic acid extraction kits and small-scale dsRNA extraction was compared. Sodium sulfite was used as an inhibitor against... -
Rapaid Screening of Apple mosaic virus in Cultivated Apples by RT-PCR
The Plant Pathology Journal 3 Pages
7 159-161 (3 pages)
A combined assay of reverse transcription and polymerase chain reaction (RT-PCR) was performed with a single pair of ApMV-specific primers and crude nucleic acid extracts from virus-infected apple for rapid detection of the virus. The PCR product was verified by restriction mapping analysis and by sequence determination. The lowest concentration of template viral RNA required for detection was 100 fg. This indicates that the RTPCR for detection of the virus is a 10(3) times more sensitive,... -
Molecular Detection and Analysis of Sweet potato feathery mottle virus from Root and Leaf Tissues of Cultivated Sweet Potato Plants
The Plant Pathology Journal 6 Pages
3 12-17 (6 pages)
of Sweet potato feathery mottle virus (SPFMV) from diseased sweet potato plants, reverse transcription and polymerase chain reaction (RT-PCR) was performed with the use of a set of virus-specific primers to amplify an 816 bp product. The viral coat protein gene was selected for the design of the primers. No PCR product was amplified when Turnip mosaic virus, Potato virus Y, or Cucumber mosaic virus were used as template in RT-PCR with the SPFMV -specific primers. The lowest concentration of...
