We describe in this report an asymmetric PCR protocol that allows specific amplification of cDNA inserts directly from ${lambda}gt11$ libraries. The method uses ${lambda}gt11$ forward or ${lambda}gt11$ reverse sequencing primer, in combination with an insert-specific primer. Asymmetric PCR conditions are crucial for specific amplification of the desired product. This protocol can potentially be extended to a wide range of PCR to optimize the reaction conditions.
