미국흰불나방 (Hyphantria cunea)의 종령유충 혈림프로 부터 저장단백질-2(SP2)을 확인한 후 salting out, cation exchange chromatography 및 gel filtration chromatography법을 이용하여 정제하고 전기영동을 통해 그 순수도를 확인하였다. SP2는 한 종류의 구성소단위 (M.W. 82.5 kDa)로 이루어져 있으며 pI값은 6.9로 측정되었고, 아미노산조성을 분석한 결과, arylphorin이 아닌 것으로 여겨진다. SP2의 N-말단서열을 조사한 결과 Hymenoptera, Diptera 및 Lepidoptera의 arylphorin과 다소 상동성이 있는 것으로 확인되었다.
Storage protein-2(SP2) of Hyphantria cunea (Lepidoptera) was confirmed from the hemolymph of the last instar larvae and purified using salting out, cation exchange chromate graphy, and gel filtration chromatography. The purity of isolated SP2 was confirmed by polyacry-lamide gel electrophoresis. SP2 is composed of single type subunit which has a molecular weight of 82.5 kDa. pI value of SP2 was determined to be 6.9. Based on the amino acid composition, SP2 is not regarded as arylphorin. SP2 has some homology in its N-terminal sequence with the arylphorins of Hymenoptera and Diptera as well as Lepidoptera.
